As far as I can tell, it means in 'at least one cult skill'. Since we're talking a *typical* rune level here, not a minimum entrance requirement, the exact number of such skills the character would have is not going to be precise - sometimes just the one, sometimes lots. OTOH, since we can assume that the character (who is, after, all, generically typical) will have concentrated on the most relevant skills, its probably a small number, although she could have quite a broad base of skills at the next bracket down.
The minimum required for rune level though, appears to be E1 in whatever skill it is you use to access the HeroPlane, and nothing more than that - any less than that and you can't do the HeroQuesting bit even at the HHD ceremonies, which would be kind of embarassing for a priest. Its being alleged that a character who only has E1 in his best skills is a bit naff for a rune level type, but even if you don't believe that, its fairly clear that most rune priests would have significantly more than the minimum requirement (season the word 'significantly' to taste).
