I think it would be cool. I'd just compare it against the opponents highest appropriate ability, Large or Tall, maybe even Strong unless I had reason to believe the NPC was meant to be short and beefy. Towers Over People would do. Apply suitable improv penalties and let the character demonstrate whether they are walking the walk or talking the talk the hard way. GM's could make a characters life hell by forcing them into a roll they are likely to lose and then needling them about it (seeing as we're into confessions of GM malice).

As for using it for other things, it depends how plausible their claim of relevance is. Being The Biggest Ant In The Hive 15w3 is surely little defense against the lowly shoe - I wouldn't let them use an ability like that unless there was a decent claim to be made. But the ability is essentially a comparative one, and I would feel that it relevant primarily when dealing with the object of the comparison.

Whats also interesting is an extenion of Bruces concern about the inflating follower - what if you have a munchkin player who with malice aforethought takes an ability like Large and deliberately sinks hero points into it? They could get quite big quite quickly.
